Both disks can be used at the same time:
The PCIE+SATA to U.2 adapter can be used at the same time, and it can be used on server motherboards that support U.2 and SATA dual protocols, but only NVME SSDs can be used with 8643 to U.2 adapter cables, and ordinary motherboards that only have a PCIE channel but not a SATA channel.
When the switch is flipped to NVME direction: NVME protocol lights up.
When the switch is flipped to the SATA direction: SATA protocol lights up.
NVME disk has NVME indicator, SATA disk has SATA indicator, this switch just switch NVME indicator to U2 connector or SATA indicator to U2 connector.
