Intact Genomics' Methionine-Auxotrophic LBA4404Met Electrocompetent Agrobacterium cells are modified to require free methionine for growth, preventing bacterial overgrowth in plant tissues during transformation. These cells are optimized for high transformation efficiency and are suitable for transgenic operations in tomatoes, tobacco, and other plants. The LBA4404Met strain carries a rifampicin resistance gene and an octopine-type Ti plasmid pAL4404Met with the vir gene. Custom aliquots are available, offering benefits such as efficient transformation systems, reduced bacterial overgrowth, and decreased antibiotic need. These electrocompetent cells, derived from A. tumefaciens, are shipped in tubes under dry ice. The kit includes the cells, pCAMBIA1391z DNA, and recovery medium, with storage at -80°C for cells, -20°C for DNA, and 4°C for the medium. Quality control ensures a transformation efficiency of 1 x 107 CFU/g pCAMBIA1391z DNA. When using these cells, handle them gently, thaw on ice, and transform immediately. Mix DNA by tapping the tube gently. To calculate transformation efficiency, transform 1 l of pCAMBIA1391z control plasmid into 50 l of cells, dilute, plate, and count colonies.
