This CLIA kit employs the Sandwich-CLIA method. It comes with a micro CLIA plate pre-coated with an antibody tailored to Rat AgRP. Samples or Standards are added to the plate's wells, combined with the specific antibody. Subsequently, a biotinylated detection antibody specific for Rat AgRP and Avidin-Horseradish Peroxidase (HRP) conjugate are added successively to each well and incubated. Unbound components are washed away. Adding the substrate solution causes luminescence in wells containing Rat AgRP, biotinylated detection antibody, and Avidin-HRP conjugate. The Relative light unit (RLU) value, measured at 425nm, correlates positively with Rat AgRP concentration. You can determine the concentration of Rat AgRP in samples by comparing the RLU value to the standard curve.
