Description

The direct capture method involves immobilizing double-stranded PCR products onto beads, which can then be easily converted into single-stranded bead-bound templates for capturing specific RNA or DNA molecules directly from solution. Alternatively, an indirect capture approach can offer faster reaction kinetics in certain cases. This indirect method allows the target sequence to be captured before being immobilized onto magnetic beads. First, a biotinylated capture-sequence (single-stranded DNA) is incubated with the sample and allowed to hybridize with the targeted RNA or DNA molecules in solution. Streptavidin-coated Dynabeads are then added to the mixture, and the hybridized sequences are immobilized onto the Dynabeads via the streptavidin-biotin bond.
