GNE-493 is a dual inhibitor targeting PI3K and mTOR (IC50s ranging from 3.4 to 32 nM). It hinders the growth of PC3 and MCF-7.1 cells (IC50s of 330 and 180 nM, respectively). In mouse models, it reduces tumor volume and lowers phosphorylated Akt, PRAS40, and S6RP levels in tumor tissue.
