Directly adding the Dynabeads-mRNA complex to RT-PCR provides advantages: mRNA elution and dilution are avoided. With mRNA bound to beads, the bead-attached oligo-dT serves as primers for first-strand cDNA synthesis, yielding a bead-bound cDNA library. This solid-phase cDNA library is particularly useful when mRNA is derived from tiny samples, with libraries generated from a single cell. The library can be regenerated and reused countless times. Dynabeads do not quench the fluorescence signal in real-time PCR but may exhibit autofluorescence, which can be compensated for by setting a Dynabeads & water background in the PCR instrument. Dynabeads can be directly used in real-time PCR instruments like ABI Prism 7000 and 7700 without affecting results. However, Dynabeads Oligo(dT) 25 are incompatible with capillary real-time PCR machines like the Roche LightCycler.
